Ashwagandha + Loratadine
Loratadine brand name: Claritin.
Ashwagandha has a minor interaction with Loratadine. Theoretically, ashwagandha might decrease the levels and clinical effects of CYP3A4 substrates.

Here is the interaction we found between these two, with the evidence behind it.
CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES
Why this applies to Loratadine
Loratadine falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Ashwagandha interacts with.
In vitro research shows that ashwagandha extract induces CYP3A4 enzymes.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the simple version: your body uses a liver enzyme called CYP3A4 to help process loratadine (Claritin). Lab studies suggest ashwagandha may speed that enzyme up a bit. If it does, your body could clear loratadine a little faster, which in theory might make it slightly less effective at calming your allergy symptoms like sneezing, runny nose, or itchy eyes.
I want to be honest about how soft this is. The concern comes only from test-tube research, not from real people, and it's rated as a minor interaction. Loratadine also isn't a prodrug, so it doesn't need this enzyme to start working. Most people taking both will be fine, but if your allergies seem harder to control after starting ashwagandha, check in with your pharmacist or doctor.

See Every Supplement That Interacts With Loratadine
This page covers Ashwagandha only. If you take other supplements — a multivitamin, a sleep aid, something for joints — the Loratadine page lists 142 supplements it has a documented interaction with, worst first, each one explained.
The most serious there right now: Activated Charcoal (major), Grapefruit (major), St. John's Wort (major), Acacia Rigidula (moderate) and Adrafinil (moderate).
By severity: 3 major · 99 moderate · 40 minor.
From our pharmacist’s overview on the Loratadine page
“Grapefruit juice is what I would flag first for anyone taking loratadine (Claritin). It blocks CYP3A4, the liver enzyme that clears the drug, so levels can climb.”
