Ayahuasca and Amlodipine Besylate, Benazepril

Here is the interaction we found between these two, with the evidence behind it.

Moderate

CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES

Why this applies to Amlodipine Besylate, Benazepril

Amlodipine Besylate, Benazepril falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Ayahuasca interacts with.

Theoretically, ayahuasca might increase plasma concentrations of CYP3A4 substrates.
Ayahuasca contains beta-carboline alkaloids. In vitro research suggests that beta-carboline alkaloids competitively inhibit CYP3A4 enzymes. This effect has not been reported in humans.
Severity Moderate Likelihood Possible Evidence D

Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.

Dr. Brian Staiger, PharmD, BCPS
Dr. Brian Staiger explains PharmD, Clinical Pharmacist

Your Lotrel is a combo pill, and the amlodipine half is cleared by a liver enzyme called CYP3A4. The beta-carboline compounds in ayahuasca can slow that enzyme down, at least in lab tests. If amlodipine clears more slowly, more of it can hang around in your blood, which could nudge its blood-pressure-lowering effect too far. That might show up as dizziness, lightheadedness, flushing, headache, or swollen ankles.

Here's the honest part: this is only seen in test-tube studies, not in real people, so it's theoretical. I still wouldn't take a chance with ayahuasca, which carries its own serious risks. Please talk with your pharmacist or doctor before mixing the two.

From our pharmacist’s overview on the Amlodipine Besylate, Benazepril page

“Grapefruit sits high on the worry list for anyone taking Lotrel (amlodipine besylate and benazepril), a two-in-one blood pressure pill.
