Here is the interaction we found between these two, with the evidence behind it.
CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES
Why this applies to Aspirin, Butalbital, Caffeine
Aspirin, Butalbital, Caffeine falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Ayahuasca interacts with.
Ayahuasca contains beta-carboline alkaloids. In vitro research suggests that beta-carboline alkaloids competitively inhibit CYP3A4 enzymes. This effect has not been reported in humans.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the gist with Fiorinal (aspirin, butalbital, caffeine) and ayahuasca. The vine in ayahuasca contains compounds that, in lab dishes, can slow down a liver enzyme called CYP3A4. Some of what's in Fiorinal, including the butalbital component, gets processed partly through that pathway. So in theory, blocking the enzyme could let these ingredients linger longer in your body and hit harder.
If that happened, you might feel more of butalbital's effects, like heavy drowsiness, dizziness, or feeling foggy and sedated. But I want to be honest: this is based only on test-tube data and has never been shown in real people. Still, ayahuasca is potent and risky on its own, so please talk with your doctor or pharmacist before combining anything with it.

“The aspirin inside Fiorinal (aspirin, butalbital, caffeine) is the reason this list deserves careful reading rather than a quick skim.
