Aspirin, Caffeine, Hydrocodone falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES — the group Ayahuasca interacts with.

Theoretically, ayahuasca might increase plasma concentrations of CYP2D6 substrates.
Ayahuasca contains beta-carboline alkaloids, including harmine and harmaline. In vitro research suggests that the beta-carboline alkaloids might inhibit CYP2D6 enzymes. This effect has not been reported in humans.

The piece of this combo product that matters here is the hydrocodone. Hydrocodone is partly a prodrug, meaning your body uses the CYP2D6 enzyme to turn some of it into hydromorphone, a stronger pain-relieving form. Ayahuasca's beta-carboline compounds (harmine and harmaline) may block CYP2D6 in lab studies, so in theory less hydrocodone gets converted, which could mean weaker pain relief for some people.

I want to be honest about the evidence: this is only seen in test-tube and animal research, not in real patients, so it's theoretical. Honestly, the bigger concern with ayahuasca is its MAOI activity mixing with other meds. Please don't change anything on your own. Talk with your pharmacist or doctor first, especially before using ayahuasca at all.

Ayahuasca contains beta-carboline alkaloids. In vitro research suggests that beta-carboline alkaloids competitively inhibit CYP3A4 enzymes. This effect has not been reported in humans.

This combo pill (Damason-P) pairs aspirin and caffeine with hydrocodone, an opioid for pain. Hydrocodone is partly handled by a liver enzyme called CYP3A4. The beta-carboline compounds in ayahuasca may block that enzyme, at least in lab studies, so in theory hydrocodone could clear a little slower and pile up. That might mean stronger effects like deep drowsiness, foggy thinking, constipation, or dangerously slow breathing.

Honestly though, this is theoretical and has only been seen in test-tube research, not in people. The bigger worry is mixing any opioid with ayahuasca's strong MAOI activity and mind-altering effects. Please don't combine these without talking to your doctor or pharmacist first.
