Sildenafil falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Ayahuasca interacts with.

Theoretically, ayahuasca might increase plasma concentrations of CYP3A4 substrates.
Ayahuasca contains beta-carboline alkaloids. In vitro research suggests that beta-carboline alkaloids competitively inhibit CYP3A4 enzymes. This effect has not been reported in humans.

Here's the gist for sildenafil: your body relies on an enzyme called CYP3A4 to break it down and clear it out. The beta-carboline compounds in ayahuasca (like harmine) may slow that enzyme down, at least in lab studies. If that happens in real life, sildenafil could hang around longer and hit harder than expected.

In practice, that might mean more of sildenafil's usual effects, things like headache, facial flushing, a stuffy nose, dizziness, or a bigger drop in blood pressure. I'll be honest, though: this is only theoretical, based on test-tube work and never confirmed in people. The bigger concern is that ayahuasca itself is risky and unpredictable. Please run any combination like this past your doctor or pharmacist first.

What Is Sildenafil Used For?

Brand name: Revatio

Sildenafil treats two distinct conditions depending on the product. Viagra (tablet) and Vybrique (dissolving oral film) are approved for erectile dysfunction (ED) — difficulty getting or keeping an erection. Generic sildenafil tablets and oral film are also approved for ED.

Revatio and several generic sildenafil products (tablets, oral suspension, and injection) are used for pulmonary arterial hypertension (PAH) — high blood pressure in the arteries of the lungs — in adults to improve exercise ability and slow the disease from getting worse. Revatio oral suspension and injection are also approved for PAH in children ages 1 to 17.
