Why this applies to Vilazodone

Vilazodone falls under SEROTONERGIC DRUGS — the group Ayahuasca interacts with.

Theoretically, ayahuasca might increase levels of serotonin, which may have additive effects with serotonergic drugs.
In vitro and animal research shows that harmaline and harmine, constituents of ayahuasca, strongly and reversibly inhibit monoamine oxidase A and weakly inhibit monoamine oxidase B. The dimethyltryptamine and tetrahydroharmine constituents of ayahuasca have also demonstrated serotonergic activity. A case of serotonin syndrome characterized by tremors, sweating, shivering, confusion, nausea, vomiting, and disorientation has been reported in a 36-year-old male who consumed 100 mL of ayahuasca while taking fluoxetine 20 mg daily. Combining serotonergic drugs with ayahuasca might increase the risk of serotonergic side effects including serotonin syndrome and cerebral vasoconstrictive disorders.

Here's the thing with Viibryd (vilazodone): it already boosts serotonin in your brain to help with depression. Ayahuasca pushes serotonin in the same direction, partly because the vine acts like an MAOI and slows the breakdown of these brain chemicals. Stacking two serotonin-raisers can be too much of a good thing.

The worry is serotonin syndrome, which can show up as shivering, sweating, tremor, a racing heart, confusion, nausea, or agitation. This concern is mostly theoretical and based on pharmacology, but a real serotonin syndrome case happened with a similar antidepressant plus ayahuasca, so it's not just hand-waving. Please don't start or stop anything on your own. Talk with your doctor or pharmacist first.

Vilazodone also falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES.

Theoretically, ayahuasca might increase plasma concentrations of CYP2D6 substrates.
Ayahuasca contains beta-carboline alkaloids, including harmine and harmaline. In vitro research suggests that the beta-carboline alkaloids might inhibit CYP2D6 enzymes. This effect has not been reported in humans.

Here's the thing I'd want you to know: Vilazodone (Viibryd) is partly cleared by a liver enzyme called CYP2D6. The beta-carboline compounds in ayahuasca (harmine and harmaline) may slow that enzyme down, at least in lab studies. If that happened in a person, your vilazodone could build up a bit, which might mean more side effects like nausea, diarrhea, dizziness, trouble sleeping, or headache.

But honestly, the bigger worry isn't the enzyme. Ayahuasca acts like an MAOI, and vilazodone already boosts serotonin. Combining the two could push serotonin dangerously high (think agitation, racing heart, sweating, confusion). That's a serious combination. Please don't mix these without talking to your doctor or pharmacist first.

Here's the thing to keep in mind with Viibryd (vilazodone): your liver uses an enzyme called CYP3A4 to clear it out. Lab studies hint that the beta-carboline compounds in ayahuasca may slow that enzyme down. If that happened in the body, vilazodone could build up and you might notice more of its usual effects, things like nausea, diarrhea, dizziness, headache, or trouble sleeping. I'll be honest, though: this CYP3A4 effect has only been seen in test-tube research, not in real people, so it's theoretical for now.

The bigger worry actually involves how both raise serotonin. Ayahuasca acts like an MAOI, and combining that with an antidepressant like vilazodone can risk a dangerous reaction called serotonin syndrome. Please don't combine these without talking to your doctor or pharmacist first.

MDD is more than just feeling sad; it's a clinical condition involving persistent low mood, loss of interest, and other symptoms that significantly affect daily life.

Vilazodone is not approved for use in children or teenagers. Its safety and effectiveness have not been established in pediatric patients.
