Guaifenesin, Hydrocodone falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES — the group Fever Bark interacts with.

Theoretically, fever bark might increase the levels and clinical effects of CYP2D6 substrates.
In vitro research suggests that yohimbine, a constituent of fever bark, inhibits CYP2D6 enzyme activity. The effects of fever bark itself are unclear.

Here's the tricky part with this combo. Your medicine pairs guaifenesin (a mucus thinner) with hydrocodone, an opioid cough and pain reliever. Hydrocodone is partly a prodrug: your body uses a liver enzyme called CYP2D6 to turn some of it into hydromorphone, a stronger active form. Fever bark contains yohimbine, which lab studies suggest can block that same enzyme.

So in theory, fever bark could slow that conversion and actually weaken hydrocodone's pain and cough relief, rather than boost it. That said, this is based only on test-tube research, and the real effect of fever bark in people is unclear. I'd honestly steer clear of fever bark on its own anyway, but please check with your pharmacist or doctor before combining them.

Fever bark is the bark of the Australian tree Alstonia constricta, used in traditional medicine for fever, diarrhea, and infections. There is very little reliable human research to support these uses, and the bark contains potent alkaloids that may cause serious side effects.
