Acetaminophen, Oxycodone falls under CNS DEPRESSANTS — the group Goldenseal interacts with.

Theoretically, goldenseal might increase the sedative effects of CNS depressants.
Goldenseal contains berberine. Animal research shows that berberine can have sedative effects. However, this effect has not been reported in humans.

Percocet pairs oxycodone, an opioid painkiller, with acetaminophen (the same ingredient in Tylenol). Oxycodone already slows down your brain a bit, which is why it can cause drowsiness, foggy thinking, and slowed breathing. The concern here is simple addition: goldenseal contains berberine, and in animal studies berberine showed mild sedative effects. So in theory, combining the two could leave you a little more sleepy or sluggish than the oxycodone alone.

I want to be honest, though: this calming effect has not actually been seen in people, so it's a theoretical caution, not a proven problem. Still, if you take Percocet and use goldenseal, mention it to your pharmacist or doctor, and watch for extra drowsiness before driving or mixing it with alcohol.

Goldenseal might increase serum levels of drugs metabolized by CYP2D6.
Clinical and in vitro research shows that goldenseal can significantly inhibit CYP2D6 enzymes, potentially increasing levels of drugs metabolized by CYP2D6.

Here's the wrinkle with this combo: the oxycodone part of your Percocet relies partly on a liver enzyme called CYP2D6 to turn it into a stronger pain-relieving form. Goldenseal can block that enzyme. So in theory, blocking CYP2D6 could shift how much active opioid you make, and the research here is decent but not airtight (lower quality human studies).

What does that mean for you day to day? You might notice your pain control feels a little off, or you could see more opioid-type effects like drowsiness, dizziness, constipation, or slowed breathing if levels climb. Note that the acetaminophen (Tylenol) portion isn't the concern here. Please don't change anything on your own. A quick chat with your pharmacist or doctor before adding goldenseal is the safe move.

Theoretically, goldenseal might increase serum levels of drugs metabolized by CYP2E1.
In vitro research shows that goldenseal root extract can inhibit the activity of CYP2E1. However, this effect has not been reported in humans.

Here's the part of Percocet that matters for this pairing: the acetaminophen (Tylenol) component. Your liver uses an enzyme called CYP2E1 to handle a small slice of acetaminophen, and that pathway is what creates a potentially toxic byproduct. In test-tube studies, goldenseal seems to slow CYP2E1 down. In theory, less activity through that route could actually mean a little less of the harmful byproduct, but honestly, this has never been shown to play out in real people.

So I wouldn't lose sleep over it. This is lab-dish evidence only, not human data. The much bigger acetaminophen concern is simply staying under the daily limit and going easy on alcohol. The oxycodone side isn't really affected here. Still, loop in your pharmacist before adding goldenseal so we can check your full list.
