Lonafarnib falls under CYTOCHROME P450 2C9 (CYP2C9) SUBSTRATES — the group Jambolan interacts with.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the quick version for Lonafarnib (Zokinvy). This medicine is partly cleared by a liver enzyme called CYP2C9. In a lab setting, jambolan appears to slow that same enzyme down. If that holds true in people, your body could clear Lonafarnib a little more slowly, letting levels creep up. Since Lonafarnib is an active drug (not a prodrug that needs switching on), higher levels usually mean a stronger effect and more side effects, things like nausea, vomiting, diarrhea, poor appetite, and fatigue.
The honest truth is that this is only test-tube evidence right now, so we can't say it will happen for sure. Eating the fruit is unlikely to matter much, but a concentrated jambolan supplement is a different story. Lonafarnib is a carefully dosed drug, so please check with your pharmacist or doctor before combining them.

Jambolan (Syzygium cumini), also called Java plum or black plum, is a tropical fruit tree traditionally used for blood sugar and digestive complaints. Human evidence for these uses is limited and not strong, so it should not replace proven treatments like diabetes medication. What Is Lonafarnib Used For?
Brand name: Zokinvy
Zokinvy (lonafarnib) is approved for patients 12 months of age and older who have a body surface area of 0.39 m² or above. It is used to reduce the risk of mortality in Hutchinson-Gilford Progeria Syndrome (HGPS) — a rare, severe premature-aging disorder.
Zokinvy is also used to treat processing-deficient Progeroid Laminopathies — a related group of rare genetic conditions — specifically those caused by a heterozygous LMNA mutation with progerin-like protein buildup, or by homozygous or compound heterozygous ZMPSTE24 mutations. It is not indicated for other Progeroid Syndromes or processing-proficient Progeroid Laminopathies.
