Amitriptyline, Perphenazine falls under ANTIPSYCHOTIC DRUGS — the group Manganese interacts with.

Theoretically, the risk for manganese toxicity might increase when taken with antipsychotic drugs.
Hallucinations and behavioral changes have been reported in a patient with liver disease who was taking haloperidol and manganese. Researchers speculate that taking manganese along with haloperidol, phenothiazine-derivatives, or other antipsychotic medications might increase the risk of manganese toxicity in some patients.

This combination pill pairs amitriptyline (an antidepressant) with perphenazine, which is an older antipsychotic. The piece that matters here is the perphenazine. The concern on file is mostly theoretical: in one patient with liver disease who was taking an antipsychotic along with manganese, doctors saw hallucinations and behavior changes, and they wondered if the manganese was building up to toxic levels.

So the worry isn't that this medicine stops working, it's that extra manganese on top of it might raise your risk of manganese buildup, which can affect the nervous system. This is weak, anecdotal evidence, and ordinary food amounts of manganese are very unlikely to cause trouble. Concentrated high-dose supplements are a different story. Please check with your pharmacist or doctor before adding a manganese supplement.
