Chlorpheniramine falls under SEIZURE THRESHOLD LOWERING DRUGS — the group Melatonin interacts with.
Some clinical evidence suggests that melatonin may increase the frequency of seizures in certain patients, particularly children with neurological disabilities.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the gist with melatonin and chlorpheniramine (Chlor-Trimeton). On paper, both can nudge the brain in a way that might make seizures a touch more likely. Chlorpheniramine is an older antihistamine that can lower the seizure threshold, and melatonin has been linked, mostly in studies of children with neurological conditions, to more frequent seizures. Put together, the worry is a slightly higher seizure risk.
I want to be honest about how soft this is. It's a theoretical concern based on how these act, not a strong, well-proven danger. For most healthy adults using both occasionally, it's unlikely to cause trouble. If you or your child has a seizure disorder, please check with your pharmacist or doctor first.
CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES
2 of 2Chlorpheniramine also falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES.
Laboratory research shows that certain lots of melatonin inhibit CYP3A4. Theoretically, combined administration of melatonin with certain drugs metabolized by CYP3A4 may reduce the metabolism of these drugs, resulting in increased serum levels. Until more is known, use melatonin cautiously in patients taking drugs metabolized by these enzymes.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the gist for these two. Some lab studies hint that melatonin may slow down an enzyme called CYP3A4, which is one of the pathways your body uses to clear chlorpheniramine. If that enzyme works a little slower, a bit more chlorpheniramine could hang around in your system. With this antihistamine, that mostly means you might feel sleepier or more groggy than usual, along with a dry mouth or a touch of dizziness.
I want to keep this in perspective, though. This is a theoretical, minor interaction based only on lab work, not on real patients. On top of that, both melatonin and chlorpheniramine can cause drowsiness on their own, so the bigger practical point is simply not to be surprised if you feel extra sleepy. If you take both regularly, just mention it to your pharmacist or doctor.

Chlorpheniramine is used to temporarily relieve common allergy symptoms caused by hay fever (allergic rhinitis) or other upper respiratory allergies. These include runny nose, sneezing, itchy or watery eyes, and itching of the nose or throat.
These uses are supported across multiple products — including tablets (such as Aller-chlor, Allergy, Allergy Relief, Allergy Time, Anti Allergy, and Chlorpheniramine Maleate), extended-release tablets (Allergy Relief and Chlorphen-12), the oral solution Ed Chlorped Jr, and the inhalers ClorRelief and ClorRelief KIDs.
