Here's the gist for Ninlaro (ixazomib). This isn't about turmeric changing how your body processes the drug. Instead, both turmeric and ixazomib can each be a little hard on the liver. Ixazomib already carries a known risk of liver problems, and there have been scattered reports of concentrated turmeric or curcumin supplements stressing the liver too. Stacking them could, in theory, add to that strain.

This is a theoretical, anecdotal concern, not a proven one. The amount of turmeric you'd sprinkle on food is very unlikely to matter. The worry is with high-dose, highly absorbable curcumin pills. Watch for yellowing skin or eyes, dark urine, belly pain, or unusual tiredness, and check with your oncologist or pharmacist before adding any turmeric supplement.

Ixazomib Citrate also falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES.

Turmeric might increase or decrease levels of drugs metabolized by CYP3A4.
In vitro and animal research show that turmeric and its constituents curcumin and curcuminoids inhibit CYP3A4. Also, 8 case reports from the World Health Organization (WHO) adverse drug reaction database describe increased toxicity in patients taking turmeric and cancer medications that are CYP3A4 substrates, including everolimus, ruxolitinib, ibrutinib, and palbociclib, and bortezomib. In another case report, a transplant patient presented with acute nephrotoxicity and elevated tacrolimus levels after consuming turmeric powder at a dose of 15 or more spoonfuls daily for ten days prior. It was thought that turmeric increased levels of tacrolimus due to CYP3A4 inhibition.
Conversely, other in vitro research suggests that turmeric induces CYP3A4 activity, leading to reduced levels of CYP3A4 substrates. An animal model suggests that induction of CYP3A4 occurs after daily curcumin use for 1 week. However, the induction of CYP3A4 by turmeric has not been reported in humans.

Here's the simple version: your body uses a liver enzyme called CYP3A4 to clear ixazomib (Ninlaro). Turmeric, mostly from its curcumin, can tinker with that same enzyme. In lab and animal studies it tends to block CYP3A4, which could let ixazomib build up and make side effects worse, think more nausea, diarrhea, low blood counts, fatigue, or nerve tingling in the hands and feet. Some lab work points the other way too, where turmeric speeds the enzyme up and weakens the drug.

Honestly, the evidence here is thin and mostly from test tubes and animals, but a few cancer-drug case reports make me cautious. Sprinkling turmeric on food is unlikely to matter; concentrated capsules are the concern. Please check with your oncology team before adding any turmeric supplement.

What Is Ixazomib Used For?

Brand name: Ninlaro

Ninlaro (ixazomib) is used to treat multiple myeloma — a cancer of plasma cells in the bone marrow. It is always used together with two other medicines, lenalidomide and dexamethasone, and is approved for adults who have already tried at least one other treatment.

Ninlaro is not recommended as maintenance therapy or for newly diagnosed multiple myeloma in combination with lenalidomide and dexamethasone outside of controlled clinical trials.
