pyrilamine, chlorpheniramine falls under Serotonergic Drugs — the group Ashwagandha interacts with.

Some animal studies have reported that ashwagandha can enhance serotonergic transmission by altering certain serotonin (5-HT) receptors. However, there is no evidence to suggest that ashwagandha increases the risk of serotonin-related effects, and there have been no published case reports of serotonin syndrome when combined with other serotonergic drugs. Nevertheless, due to the lack of extensive studies on the matter and the fact that ashwagandha appears to affect serotonergic pathways, it would be prudent to exercise caution when combining it with drugs that affect serotonin. If you're using a product like Derma-Pax that contains pyrilamine and chlorpheniramine, here's the short version: these are older antihistamines, and the concern with ashwagandha is fairly small. Animal studies hint that ashwagandha may nudge serotonin pathways a bit, but there are no real-world reports of problems when it's paired with serotonin-affecting medicines. This is a theoretical, minor interaction backed by weak evidence.

What's more worth keeping an eye on is that both antihistamines and ashwagandha can make you feel drowsy and relaxed. Together they might leave you a little sleepier or more foggy than usual. Nothing dangerous for most healthy adults, but go easy on driving until you know how you feel, and check with your pharmacist if you have questions.
