Aspirin, Codeine Phosphate, Phenobarbital falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES — the group Ayahuasca interacts with.

Theoretically, ayahuasca might increase plasma concentrations of CYP2D6 substrates.
Ayahuasca contains beta-carboline alkaloids, including harmine and harmaline. In vitro research suggests that the beta-carboline alkaloids might inhibit CYP2D6 enzymes. This effect has not been reported in humans.

Here's the wrinkle with this combo product: the codeine part is a prodrug. Your body uses an enzyme called CYP2D6 to convert codeine into morphine, which is what actually relieves pain. Ayahuasca's beta-carboline compounds (harmine and harmaline) may block that same enzyme. So in theory, less codeine would be turned into morphine, meaning weaker pain relief rather than more side effects. The aspirin and phenobarbital pieces aren't really driven by CYP2D6, so they're less of a concern here.

Honestly, this is based on lab studies only and hasn't been shown in real people. The bigger issue is that ayahuasca acts as an MAOI and mixing it with opioids and a barbiturate can be genuinely dangerous. Please talk with your pharmacist or doctor before combining these.
