Methylprednisolone falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Ayahuasca interacts with.

Theoretically, ayahuasca might increase plasma concentrations of CYP3A4 substrates.
Ayahuasca contains beta-carboline alkaloids. In vitro research suggests that beta-carboline alkaloids competitively inhibit CYP3A4 enzymes. This effect has not been reported in humans.

Here's the short version: your body relies on a liver enzyme called CYP3A4 to help clear methylprednisolone (Medrol). Ayahuasca contains beta-carboline compounds that, in lab studies, appear to slow that same enzyme down. If that happened in a real person, methylprednisolone could linger longer and build up, which might mean stronger steroid effects and more side effects like trouble sleeping, mood swings, jittery energy, higher blood sugar, fluid retention, or stomach upset.

The honest part: this is only a theoretical concern based on test-tube work, never shown in people. I'm not raising alarms here. Still, ayahuasca carries serious risks of its own, so please loop in your doctor or pharmacist before combining anything.

What Is Methylprednisolone Used For?

Brand names: Medrol, Medrol Dosepak

Methylprednisolone tablets (Medrol, Methylpred DP) are used to treat a broad set of conditions including endocrine disorders, rheumatic and joint conditions, autoimmune diseases such as systemic lupus erythematosus, severe allergic states, skin diseases, eye inflammation, breathing disorders like asthma, blood and kidney diseases, and certain cancers.

Methylprednisolone acetate injection (Depo-Medrol and others) is used by intramuscular injection when oral therapy isn't feasible, and can also be given directly into joints, soft tissue, or lesions to treat local inflammation from conditions such as rheumatoid arthritis, bursitis, and skin disorders. The injection is not approved for intrathecal (spinal) use.
