Tadalafil falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Ayahuasca interacts with.

Theoretically, ayahuasca might increase plasma concentrations of CYP3A4 substrates.
Ayahuasca contains beta-carboline alkaloids. In vitro research suggests that beta-carboline alkaloids competitively inhibit CYP3A4 enzymes. This effect has not been reported in humans.

Here's the basic idea: your body relies on a liver enzyme called CYP3A4 to clear tadalafil out of your system. The beta-carboline compounds in ayahuasca (like harmine) may block that enzyme. In lab dish studies, this slowed CYP3A4 down, which in theory could let tadalafil build up higher than usual and linger longer.

If that happened, you might notice more of tadalafil's typical effects, such as headache, facial flushing, nasal congestion, indigestion, or a bigger drop in blood pressure causing dizziness. I'll be honest though, this is only seen in test-tube research, not in real people, so it stays theoretical for now. Ayahuasca also carries its own serious risks, so please loop in your doctor or pharmacist before mixing anything.

About Tadalafil

What Is Tadalafil Used For?

Brand names: Cialis, Tadliq

Tadalafil tablets (including Cialis and generic tadalafil) are used to treat erectile dysfunction (ED), the symptoms of benign prostatic hyperplasia (BPH — an enlarged prostate), or both conditions together. When used with the drug finasteride to start BPH treatment, this combination is recommended for up to 26 weeks.

Adcirca, Alyq (tablets), and Tadliq (oral suspension) are indicated specifically for pulmonary arterial hypertension (PAH — high blood pressure in the lungs), to improve the ability to exercise.
