Why this applies to Venlafaxine

Venlafaxine falls under SEROTONERGIC DRUGS — the group Ayahuasca interacts with.

Theoretically, ayahuasca might increase levels of serotonin, which may have additive effects with serotonergic drugs.
In vitro and animal research shows that harmaline and harmine, constituents of ayahuasca, strongly and reversibly inhibit monoamine oxidase A and weakly inhibit monoamine oxidase B. The dimethyltryptamine and tetrahydroharmine constituents of ayahuasca have also demonstrated serotonergic activity. A case of serotonin syndrome characterized by tremors, sweating, shivering, confusion, nausea, vomiting, and disorientation has been reported in a 36-year-old male who consumed 100 mL of ayahuasca while taking fluoxetine 20 mg daily. Combining serotonergic drugs with ayahuasca might increase the risk of serotonergic side effects including serotonin syndrome and cerebral vasoconstrictive disorders.

Here's the concern with mixing ayahuasca and venlafaxine (Effexor). Venlafaxine already raises serotonin levels in your brain as part of how it treats depression and anxiety. Ayahuasca pushes serotonin up too, partly because the vine acts like an MAO inhibitor and partly from its own serotonin-like compounds. Stacking both can drive serotonin too high.

When serotonin climbs too far, you can get serotonin syndrome, with shivering, sweating, a racing heart, tremor, confusion, nausea, and agitation. That can become a medical emergency. A real case of this was reported with a similar antidepressant. The evidence here is mostly theoretical and from lab work, but the risk is taken seriously. Please don't combine these without first talking to your doctor or pharmacist.

Venlafaxine also falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES.

Theoretically, ayahuasca might increase plasma concentrations of CYP2D6 substrates.
Ayahuasca contains beta-carboline alkaloids, including harmine and harmaline. In vitro research suggests that the beta-carboline alkaloids might inhibit CYP2D6 enzymes. This effect has not been reported in humans.

Here's the thing I'd want you to know about combining ayahuasca with venlafaxine (Effexor). Your liver uses an enzyme called CYP2D6 to help clear venlafaxine. Lab studies suggest the beta-carboline compounds in ayahuasca (harmine and harmaline) may block that enzyme, which in theory could let more venlafaxine build up in your body. That might mean stronger side effects like nausea, jitteriness, a racing heart, high blood pressure, sweating, or trouble sleeping.

But honestly, my bigger worry is a different one. Ayahuasca acts like an MAOI, and venlafaxine already raises serotonin. Together they can push serotonin dangerously high (serotonin syndrome), which can be a medical emergency. Please talk to your doctor or pharmacist before mixing these.

Venlafaxine is used to treat major depressive disorder (MDD) across all available formulations. Extended-release capsules (including Effexor XR) are also approved for generalized anxiety disorder (GAD), social anxiety disorder (SAD), and panic disorder (PD).

Extended-release tablets are approved for major depressive disorder and social anxiety disorder. Immediate-release venlafaxine tablets are indicated for major depressive disorder only.
