Why this applies to Vortioxetine

Vortioxetine falls under SEROTONERGIC DRUGS — the group Ayahuasca interacts with.

Theoretically, ayahuasca might increase levels of serotonin, which may have additive effects with serotonergic drugs.
In vitro and animal research shows that harmaline and harmine, constituents of ayahuasca, strongly and reversibly inhibit monoamine oxidase A and weakly inhibit monoamine oxidase B. The dimethyltryptamine and tetrahydroharmine constituents of ayahuasca have also demonstrated serotonergic activity. A case of serotonin syndrome characterized by tremors, sweating, shivering, confusion, nausea, vomiting, and disorientation has been reported in a 36-year-old male who consumed 100 mL of ayahuasca while taking fluoxetine 20 mg daily. Combining serotonergic drugs with ayahuasca might increase the risk of serotonergic side effects including serotonin syndrome and cerebral vasoconstrictive disorders.

Here's the concern with mixing these two. Vortioxetine (Trintellix) works by raising serotonin in your brain. Ayahuasca pushes serotonin up too, partly because compounds in the vine act like MAO inhibitors, which slow the breakdown of serotonin. Stacking both means serotonin can climb higher than your body wants.

When serotonin gets too high you can develop serotonin syndrome, with symptoms like shivering, sweating, fast heartbeat, agitation, tremor, nausea, confusion, and in severe cases dangerous changes in blood pressure or temperature. This warning is based on pharmacology and one reported case with a similar antidepressant, so it's theoretical but plausible, not guaranteed. Please don't change anything on your own. Talk with your doctor or pharmacist first, since combining these is genuinely risky.

Vortioxetine also falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES.

Theoretically, ayahuasca might increase plasma concentrations of CYP2D6 substrates.
Ayahuasca contains beta-carboline alkaloids, including harmine and harmaline. In vitro research suggests that the beta-carboline alkaloids might inhibit CYP2D6 enzymes. This effect has not been reported in humans.

Here's the thing to know about pairing these two. Your body relies heavily on an enzyme called CYP2D6 to clear vortioxetine (Trintellix) out of your system. Ayahuasca contains compounds (harmine and harmaline) that, in lab studies, appear to block that same enzyme. If that blocking happened in real life, your vortioxetine could build up and hit harder, meaning more nausea, dizziness, headache, or trouble sleeping.

But honestly, the bigger worry here is a different one: ayahuasca also acts like an MAOI, and combining an MAOI with a serotonin medication like vortioxetine can trigger dangerous serotonin syndrome. That's a serious safety concern, not just a theoretical one. Please talk with your doctor or pharmacist before going anywhere near this combination.
