Tazemetostat Hydrobromide falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Cascara Sagrada interacts with.

Theoretically, cascara sagrada might decrease the effects of CYP3A4 substrates.
In vitro research suggests that cascara sagrada can induce CYP3A4 enzymes, albeit to a much lower degree than rifampin, a known CYP3A4 inducer.

Here's the basic idea. Your body clears tazemetostat (Tazverik) using a liver enzyme called CYP3A4. Lab studies hint that cascara sagrada may gently speed up that same enzyme, a bit like a very weak version of strong inducers. If that happened in real life, your body could break tazemetostat down a little faster, which in theory might lower its blood levels and make your cancer treatment work less well over time.

I want to be honest about how soft this signal is. It comes only from test-tube research, the effect was small, and it's rated minor. Still, because tazemetostat is an important cancer drug, don't add cascara on your own. Check with your oncology team or pharmacist first.

Cascara sagrada is a stimulant laxative made from the aged bark of a Pacific Northwest tree, used mainly for short-term relief of constipation. Because it can cause cramping, dehydration, and electrolyte problems—and was removed from over-the-counter laxative products in the U.S. due to safety concerns—it should only be used briefly, if at all, and with guidance from a pharmacist or doctor.

About Tazemetostat

What Is Tazemetostat Used For?

Brand name: Tazverik

Tazverik (tazemetostat) tablets are used to treat adults and patients aged 16 and older with metastatic or locally advanced epithelioid sarcoma (a rare soft-tissue cancer) that cannot be completely removed by surgery.

Tazverik is also used in adults with relapsed or refractory follicular lymphoma — meaning the lymphoma has come back or stopped responding to treatment — either when the tumor carries a specific EZH2 gene mutation (confirmed by an FDA-approved test) after at least two prior treatments, or when there are no other satisfactory treatment options. Both uses were approved based on response rates, and continued approval may depend on results from ongoing confirmatory studies.
