Adagrasib falls under HEPATOTOXIC DRUGS — the group Epistane interacts with.

Theoretically, concomitant use might have additive adverse hepatotoxic effects.
Epistane is an anabolic androgenic steroid, and more specifically a 17-alpha-alkylated androgen. Therefore, there is concern that epistane has hepatotoxic effects. Also, epistane is structurally similar to epitiostanol, which has been associated with hepatotixicity in a case series of four patients.

Here's the heart of it: Adagrasib (Krazati) is a cancer medication that can be tough on your liver on its own. During treatment, your oncology team watches your liver enzymes closely, because Adagrasib can sometimes cause them to climb. Epistane is a methylated anabolic steroid that is also known to stress the liver. Stacking two liver-stressing substances could, in theory, add up and raise your risk of liver injury.

To be fair, this concern is theoretical, based on how these two behave rather than reports of them being combined. Still, with a serious medication like Adagrasib, I wouldn't take chances. Epistane isn't a safe supplement to begin with, and I'd strongly urge you to talk with your oncologist or pharmacist before going anywhere near it.

Epistane is a synthetic anabolic steroid (a methylated 'designer steroid'), not a true dietary supplement or herb, even though it has been sold as a 'prohormone.' It is liver-toxic, can disrupt your natural hormones, and is illegal to sell as a supplement in the US under the Designer Anabolic Steroid Control Act. About Adagrasib

What Is Adagrasib Used For?

Brand name: Krazati

Krazati (adagrasib) is used to treat adults with locally advanced or metastatic non-small cell lung cancer (NSCLC) that carries a specific genetic change called a KRAS G12C mutation. It is used as a single agent for NSCLC after at least one prior treatment, and patients must be confirmed to have this mutation by an FDA-approved test.

Krazati is also used in combination with another drug called cetuximab to treat adults with KRAS G12C-mutated locally advanced or metastatic colorectal cancer (CRC) who have already been treated with specific prior chemotherapy regimens. Both uses were approved under the FDA's accelerated approval program based on tumor response data, and continued approval may depend on results from confirmatory trials.
