Why this applies to Elagolix Sodium

Elagolix Sodium falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Gymnema interacts with.

Theoretically, gymnema might increase levels of drugs metabolized by CYP3A4.
One in vitro study using rat liver microsomes shows that gymnema can modestly inhibit the CYP3A4 enzyme. However, other in vitro research using human liver microsomes shows that gymnema does not affect CYP3A4 activity. Animal research also shows that gymnema does not alter the function of CYP3A4. In one study in rats, oral administration of gymnema for 7 days did not alter the clearance of amlodipine, a CYP3A4 substrate.

Here's the short version on combining gymnema with Orilissa (elagolix). Your body clears elagolix partly through a liver enzyme called CYP3A4. The thinking is that gymnema might slow that enzyme down a little, which could let elagolix build up slightly and, in theory, nudge up its common effects like hot flashes, night sweats, headache, nausea, or mood changes.

The good news is that this concern rests on very weak evidence. It comes mostly from a single test-tube study using rat tissue, while studies in human liver tissue and live animals showed no real effect on CYP3A4. So a meaningful interaction is unlikely. Still, since elagolix already affects your hormones, it's smart to check with your pharmacist or doctor before adding gymnema.

Orilissa (elagolix) is used to treat moderate to severe pain associated with endometriosis — a condition where tissue similar to the uterine lining grows outside the uterus, causing significant pain.

This includes general pelvic pain as well as pain during sex (dyspareunia). How long you can take it depends on your dose and your overall health, so your prescriber will tailor the treatment duration to you.
