N,n-dmpea + Methadone
Methadone brand name: Dolophine.
N,n-dmpea has a moderate interaction with Methadone. In vitro research shows that N,N-DMPEA strongly inhibits cytochrome P450 2D6 (CYP2D6).

Here is the interaction we found between these two, with the evidence behind it.
CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES
Why this applies to Methadone
Methadone falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES — the group N,n-dmpea interacts with.
Some drugs metabolized by CYP2D6 include amitriptyline (Elavil), clozapine (Clozaril), codeine, desipramine (Norpramin), donepezil (Aricept), fentanyl (Duragesic), flecainide (Tambocor), fluoxetine (Prozac), meperidine (Demerol), methadone (Dolophine), metoprolol (Lopressor, Toprol XL), olanzapine (Zyprexa), ondansetron (Zofran), tramadol (Ultram), trazodone (Desyrel), and others.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the gist for methadone. Lab tests suggest N,N-DMPEA may block an enzyme called CYP2D6, which is one of the pathways your body uses to clear methadone. If that pathway slows down, methadone could build up a bit more than usual, and that can mean stronger opioid effects like heavy drowsiness, slowed or shallow breathing, and constipation.
I want to be honest about how shaky this evidence is: it comes only from test-tube studies, not real people, so we can't say how big the effect would be in you. But methadone is a powerful, long-acting opioid where even small changes matter, and these stimulant-type supplements are poorly studied. Please loop in your doctor or pharmacist before mixing the two.

- ProbableInteraction has not been documented in well-controlled studies, however, the interaction has been demonstrated in some small human studies or in controlled animal studies in conjunction with multiple case reports.
- PossibleInteraction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
- UnlikelyInteraction has been demonstrated in animal or in lab research but has been shown not to occur in humans.

From our pharmacist’s overview on the Methadone page
“If you take methadone, choose your supplements with genuine care. Of the 223 ingredients on this page, most either add to methadone's sedating effect or change how fast your body clears it, which can push its blood level up or down.”

About N,n-dmpea
What is N,n-dmpea?
- Mood or energy support
- Pre-workout or stimulant effect
- Cognitive or focus support
N,N-DMPEA (N,N-dimethylphenethylamine) is a synthetic relative of phenethylamine sometimes marketed in stimulant or pre-workout supplements. There is very little reliable human research on its effects or safety, and it is not a well-established dietary ingredient, so it should be approached with real caution.
About Methadone
What Is Methadone Used For?
Brand name: Dolophine
Methadone treats two main problems. Some products are used for severe, persistent pain that needs daily, around-the-clock opioid treatment when other options are not adequate. Some products are used for opioid addiction (heroin or other morphine-like drugs), as detoxification or as maintenance treatment along with social and medical services.
Not every product is approved for every use. Methadone tablets, oral solution and oral concentrate (Intensol) list both pain and opioid addiction. Diskets, Methadose Dispersible tablets and the Methadose and Methadone Hydrocloride oral concentrates are labeled only for opioid addiction. Methadone injection is for severe pain, and for short-term treatment of opioid dependence in people who cannot take medicine by mouth, such as hospitalized patients. It is not approved for outpatient opioid dependence treatment. Methadone is not meant to be used as an as-needed pain reliever.
