Why this applies to Imipramine

Imipramine falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES — the group N,n-dmpea interacts with.

In vitro research shows that N,N-DMPEA strongly inhibits cytochrome P450 2D6 (CYP2D6). Theoretically, concomitant use with drugs metabolized by CYP2D6 might increase the risk for adverse effects from these drugs.
Some drugs metabolized by CYP2D6 include amitriptyline (Elavil), clozapine (Clozaril), codeine, desipramine (Norpramin), donepezil (Aricept), fentanyl (Duragesic), flecainide (Tambocor), fluoxetine (Prozac), meperidine (Demerol), methadone (Dolophine), metoprolol (Lopressor, Toprol XL), olanzapine (Zyprexa), ondansetron (Zofran), tramadol (Ultram), trazodone (Desyrel), and others.

Here's the gist for imipramine (Tofranil). Your liver uses an enzyme called CYP2D6 to help break this medicine down and clear it from your body. Lab studies suggest N,N-DMPEA can block that same enzyme. If clearance slows, imipramine levels could creep up, which may mean stronger side effects like a dry mouth, constipation, drowsiness, dizziness on standing, or a faster or irregular heartbeat.

I want to be honest with you: this is theoretical, based only on test-tube work, not real people. Still, tricyclics like imipramine have a narrow safe range, so even small shifts matter. On top of that, N,N-DMPEA itself is poorly studied and a bit of a wild card. Please check with your pharmacist or doctor before combining them.

What Is Imipramine Used For?

Brand names: Tofranil, Tofranil PM

Imipramine is used to relieve symptoms of depression. Both Imipramine Hydrochloride tablets and Imipramine Pamoate capsules are approved for this use. Endogenous depression — the kind that seems to arise from within rather than a clear outside cause — tends to respond especially well. It may take one to three weeks before you notice the full benefit.

Imipramine Hydrochloride tablets are also approved as a short-term, add-on treatment to help reduce nighttime bedwetting (nocturnal enuresis) in children aged 6 and older, after a doctor has ruled out physical causes.
