Why this applies to Propoxyphene

Propoxyphene falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES — the group N,n-dmpea interacts with.

In vitro research shows that N,N-DMPEA strongly inhibits cytochrome P450 2D6 (CYP2D6). Theoretically, concomitant use with drugs metabolized by CYP2D6 might increase the risk for adverse effects from these drugs.
Some drugs metabolized by CYP2D6 include amitriptyline (Elavil), clozapine (Clozaril), codeine, desipramine (Norpramin), donepezil (Aricept), fentanyl (Duragesic), flecainide (Tambocor), fluoxetine (Prozac), meperidine (Demerol), methadone (Dolophine), metoprolol (Lopressor, Toprol XL), olanzapine (Zyprexa), ondansetron (Zofran), tramadol (Ultram), trazodone (Desyrel), and others.

Here's the gist for propoxyphene (Darvon). Your body relies partly on an enzyme called CYP2D6 to process this opioid pain reliever. Lab studies suggest N,N-DMPEA can strongly block that same enzyme. If it does the same thing in people, propoxyphene could clear more slowly and build up in your system, which may mean stronger sedation, dizziness, constipation, or slowed breathing.

I want to be straight with you, though: this concern comes only from test-tube research, not real human studies, and we honestly know very little about N,N-DMPEA's safety to begin with. Since propoxyphene is a potent opioid where small shifts matter, please don't pair these without first checking with your pharmacist or doctor.

This three-ingredient combination — butalbital, acetaminophen, and caffeine — is used to relieve the symptoms of tension headache (also called muscle contraction headache). All supported products, including tablets, capsules, and oral solution, share this same approved use.

It is worth knowing that evidence for using this combination to treat repeated, frequent tension headaches over time is limited. Because butalbital can be habit-forming, extended or repeated use is not recommended.
