Colchicine, Probenecid falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Padang Cassia interacts with.

In vitro research shows that a methanol extract of Padang cassia inhibits cytochrome P450 3A4 (CYP3A4) activity. Theoretically, taking Padang cassia with CYP3A4 substrates might increase the effects and side effects of these substrates.

Here's the gist for ColBenemid (colchicine plus probenecid). The colchicine part is cleared from your body partly by an enzyme called CYP3A4. In a lab study, Padang cassia (a cassia cinnamon) appeared to slow that enzyme down. If that held true in people, colchicine could clear more slowly and build up, which can mean stronger effects and more side effects like nausea, stomach cramps, diarrhea, or in rare cases muscle weakness.

I want to keep this in perspective. This is based only on a test-tube study, not real patients, so it's theoretical. Colchicine does have a narrow safety window, so I'd be cautious with concentrated cinnamon supplements. Sprinkling cinnamon on your food is very unlikely to matter. Before adding a cinnamon supplement, just run it by your pharmacist or doctor.

Padang Cassia is a type of cinnamon (Indonesian or Korintje cinnamon) grown mainly in Indonesia and used widely as a kitchen spice. As a supplement, evidence for health benefits like blood sugar control is limited and mixed, and like other cassia cinnamons it naturally contains coumarin, which can be harmful to the liver in large amounts.
