Aminoglutethimide falls under CNS DEPRESSANTS — the group Tianeptine interacts with.

Tianeptine has sedative effects; high doses may have additive effects with other CNS depressants.
Higher doses of tianeptine have opioid like effects and may have additive effects with other sedative drugs. In one case report, a 42-year-old male presented with respiratory depression and aspiration pneumonia due to concomitant use of high-dose tianeptine and alprazolam.

Here's the short version: this is about added drowsiness, not a chemical clash inside your liver. Aminoglutethimide (Cytadren), used to lower hormone production in certain cancers and Cushing's syndrome, can already make people feel sleepy, dizzy, and unsteady, especially when you first start it. Tianeptine, when taken in the high doses found in those over-the-counter 'gas station' products, behaves like an opioid and can also slow you down.

Stack the two and those sedating effects can pile on top of each other. In a worst case, that means heavy drowsiness, confusion, or even slowed breathing. This concern is based on how these drugs act, not on solid studies with this exact pair, so think of it as a caution. Honestly, I'd steer clear of OTC tianeptine entirely. Please loop in your doctor or pharmacist before combining anything.
