This product contains 2 active ingredients: SAMe and folate. SAMe (S-adenosylmethionine) is a compound your body makes naturally; it's involved in mood, joint health, and liver function.
Folate is a B vitamin that supports cell division and energy metabolism. The capsules also contain inactive ingredients — vegetable capsule material, microcrystalline cellulose (a binder), silicon dioxide, and magnesium stearate — that help hold everything together and keep it stable on the shelf.
Does it work?
Strong evidence
SAMe is likely effective for osteoarthritis, helping reduce joint pain and stiffness. It's possibly effective for depression and for cholestasis (a liver condition involving reduced bile flow).
For Alzheimer disease and ADHD, the evidence isn't established enough in our data to rate it. If you're considering SAMe for one of these conditions, talk with your doctor about whether it fits your treatment plan.
How safe is it?
Well-documented data
SAMe is generally well tolerated in typical doses, though side effects become more common at higher doses. The most common ones are stomach upset — nausea, diarrhea, constipation, or vomiting — along with headache, dizziness, insomnia, dry mouth, and nervousness.
Some people also report rash, itching, or hair loss. Important caution: SAMe may trigger mania in people with bipolar disorder, and safety is not well established for long-term use — use only under your doctor's or pharmacist's guidance.

Before taking SAMe, check with your doctor or pharmacist if you take serotonergic drugs — that includes most antidepressants (SSRIs, SNRIs, tricyclics), certain pain medications, and other serotonin-affecting drugs — because of the Moderate risk of serotonin syndrome. Also double-check if you take levodopa for Parkinson disease, as SAMe may reduce its effectiveness.

Theoretically, combining serotonergic drugs with SAMe might increase the risk of serotonergic side effects, including serotonin syndrome and cerebral vasoconstrictive disorders. In one case report, SAMe 100 mg intramuscularly was given daily with clomipramine (Anafranil) 25 mg per day. When the clomipramine dose was increased to 75 mg per day the patient experienced serotonin syndrome about 48-72 hours later, requiring hospitalization.
