Here's the gist for this combo. Adagrasib (Krazati) blocks an enzyme in your liver called CYP3A4. That same enzyme is what breaks down modafinil, the active stimulant your body makes after taking adrafinil. So if you take adrafinil while on Adagrasib, the modafinil may hang around longer and hit harder than expected.

In plain terms, you could feel more stimulant-like effects: trouble sleeping, jitteriness, a racing heart, headache, or anxiety. This concern is theoretical, based on how the drugs are processed rather than real-world reports, so it's not a sound-the-alarm situation. Still, adrafinil is unregulated and rough on the liver. Please check with your pharmacist or oncologist before pairing it with Adagrasib.

Here's the gist for Adagrasib (Krazati), your cancer-fighting pill. Once adrafinil reaches your liver, it turns into modafinil, and modafinil can gently speed up a liver enzyme called CYP3A4. That same enzyme helps break down Adagrasib. So in theory, pairing the two could clear Adagrasib out of your body a little faster, which might leave you with lower drug levels and a weaker effect against the cancer. With a targeted therapy like this, you really want steady, reliable levels.

I'll be honest: this is a theoretical concern based on how the drugs work, not something proven in studies, and modafinil is only a weak inducer. Still, please don't combine them without first checking with your oncologist or pharmacist.

About Adagrasib

What Is Adagrasib Used For?

Brand name: Krazati

Krazati (adagrasib) is used to treat adults with locally advanced or metastatic non-small cell lung cancer (NSCLC) that carries a specific genetic change called a KRAS G12C mutation. It is used as a single agent for NSCLC after at least one prior treatment, and patients must be confirmed to have this mutation by an FDA-approved test.

Krazati is also used in combination with another drug called cetuximab to treat adults with KRAS G12C-mutated locally advanced or metastatic colorectal cancer (CRC) who have already been treated with specific prior chemotherapy regimens. Both uses were approved under the FDA's accelerated approval program based on tumor response data, and continued approval may depend on results from confirmatory trials.
