Here's the gist for this pairing. Amprenavir (Agenerase), an HIV protease inhibitor, is a strong blocker of a liver enzyme called CYP3A4. That same enzyme helps clear modafinil, which is what your body turns adrafinil into. So when these two are around together, the modafinil from adrafinil may not be broken down as quickly and could build up more than expected.

In real life, that might mean stronger stimulant effects: trouble sleeping, a racing or pounding heart, jitteriness, headache, or higher blood pressure. Keep in mind this is a theoretical concern based on how the drugs work, not hard proof. Adrafinil is also unregulated and tough on the liver. Here's the gist: your body turns adrafinil into modafinil, and modafinil can gently speed up a liver enzyme called CYP3A4. Amprenavir (Agenerase), an HIV protease inhibitor, is broken down by that very same enzyme. So in theory, taking adrafinil could nudge your body to clear amprenavir a little faster, which might lower its blood levels.

Why does that matter? With HIV medicines, keeping levels steady is important. If amprenavir drops too low, the virus has more room to slip through, and that can lead to weaker control or resistance over time. This concern is theoretical and based on how the drugs work, not on real-world reports, and modafinil is only a weak inducer. Still, please loop in your doctor or pharmacist before combining them.
