Theoretically, adrafinil might increase the levels and clinical effects of CYP2C9 substrates.
Adrafinil is a precursor to the stimulant prescription drug modafinil. Modafinil has been shown to reduce CYP2C9 expression in vitro.

Here's the gist with Seglentis (the combo pill of celecoxib plus tramadol). Adrafinil turns into modafinil in your body, and in lab studies modafinil can slightly slow down an enzyme called CYP2C9. Celecoxib leans on CYP2C9 to be cleared, so in theory it could build up a bit. That might mean more of the usual celecoxib side effects, like stomach upset, heartburn, fluid retention, higher blood pressure, or, less commonly, kidney strain.

I want to be honest, though: this is a theoretical concern based only on test-tube data, not real patients, so it may not amount to much in practice. Since adrafinil is unregulated and hard on the liver, please run this combo by your doctor or pharmacist before mixing them.

Seglentis is a combo of two medicines: celecoxib (an anti-inflammatory pain reliever) and tramadol (an opioid pain medicine). Adrafinil turns into modafinil in your body, and modafinil can mildly speed up a liver enzyme called CYP3A4 that helps process both of these drugs. In theory, that faster clearance could lower their levels and leave you with less pain relief than you expect.

Here's an interesting wrinkle with tramadol: it's a prodrug that needs activation to work fully, so the picture is mixed and not fully predictable. Honestly, this concern is theoretical, based on how the drugs behave on paper, not on real patient reports. It's worth a quick chat with your pharmacist or doctor, but no need to panic.
