Here's the gist for this pairing. Encorafenib (Braftovi) can mildly block a liver enzyme called CYP3A4. That same enzyme is what breaks down modafinil, the active stimulant your body makes from adrafinil. So in theory, taking encorafenib could slow modafinil's breakdown and let it build up, which may mean stronger stimulant effects like jitteriness, a racing heart, trouble sleeping, headache, or higher blood pressure.

I'll be honest, this is a theoretical concern based on how the drugs are processed, not something proven in real patients. Still, encorafenib is a serious cancer medicine, and adrafinil is unregulated and tough on the liver. Please don't add it without first checking with your oncology team or pharmacist.

Here's the gist for Encorafenib (Braftovi). Your liver turns adrafinil into modafinil, and modafinil can mildly speed up an enzyme called CYP3A4. Encorafenib is cleared partly through that same enzyme, so in theory adrafinil could nudge your body into breaking it down a bit faster. That could mean lower encorafenib levels and, potentially, less of the cancer-fighting benefit you're counting on.

I want to be honest about the strength of this one: it's theoretical, based on how these drugs behave rather than real-world cases. Still, with a serious cancer therapy, you don't want to risk weakening it. Adrafinil is also unregulated and tough on the liver, which matters here too. Please loop in your oncologist or pharmacist before combining them.

About Encorafenib

What Is Encorafenib Used For?

Brand name: Braftovi

Braftovi is used to treat cancers that have a specific change (mutation) in a gene called BRAF — specifically the V600E or V600K mutation, which must be confirmed by an FDA-authorized test before starting treatment. It is always used in combination with other medicines, never alone as standard therapy.

Braftovi (in combination with binimetinib) treats unresectable or metastatic melanoma with a BRAF V600E or V600K mutation. For metastatic colorectal cancer (mCRC) with a BRAF V600E mutation, Braftovi is used either with cetuximab and fluorouracil-based chemotherapy, or with cetuximab alone after prior therapy. Braftovi (in combination with binimetinib) also treats metastatic non-small cell lung cancer (NSCLC) with a BRAF V600E mutation.
