Linagliptin falls under CYTOCHROME P450 3A4 (CYP3A4) INHIBITORS — the group Adrafinil interacts with.

Theoretically, drugs that inhibit CYP3A4 might increase the levels and clinical effects of adrafinil.
Adrafinil is a precursor to the stimulant prescription drug modafinil. Modafinil is metabolized by CYP3A4.

Here's the key thing about this pairing: the concern is mostly the other way around. Your wakefulness supplement, adrafinil, turns into modafinil in the body, and modafinil is broken down by an enzyme called CYP3A4. Linagliptin (Tradjenta) is a mild blocker of that same enzyme, so in theory it could slow the breakdown of modafinil and let a little more build up. That might mean stronger stimulant effects like jitteriness, a faster heartbeat, trouble sleeping, or feeling on edge.

I want to be honest about how strong this evidence is: it's theoretical, based on how the drugs work on paper, not on real patient reports. Linagliptin is also only a weak CYP3A4 inhibitor, so any effect is likely small. Still, since adrafinil is unregulated and hard on the liver, please check with your doctor or pharmacist before combining them.
