Trazodone falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Adrafinil interacts with.

Theoretically, adrafanil might reduce levels and clinical effects of CYP3A4 substrates.
Adrafinil is a precursor to the stimulant prescription drug modafinil. Modafinil has been shown to weakly induce CYP3A4 activity.

Here's the gist of it. When your body turns adrafinil into modafinil, that modafinil can mildly speed up an enzyme in your liver called CYP3A4. Since trazodone is partly broken down by that same enzyme, taking the two together could nudge your trazodone levels a little lower than usual.

If that happens, your trazodone might not work quite as well, so you could notice a return of low mood or, since many folks take it for sleep, trouble falling or staying asleep. I want to be straight with you though: this is a theoretical concern based on how the drugs work, not strong proof, and the effect on CYP3A4 is considered weak. Adrafinil also carries its own liver risks, so please run this combo by your doctor or pharmacist first.
