Eszopiclone falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Cascara Sagrada interacts with.

Theoretically, cascara sagrada might decrease the effects of CYP3A4 substrates.
In vitro research suggests that cascara sagrada can induce CYP3A4 enzymes, albeit to a much lower degree than rifampin, a known CYP3A4 inducer.

Your body relies on a liver enzyme called CYP3A4 to clear eszopiclone (Lunesta) out of your system. Lab studies hint that cascara sagrada might gently nudge that enzyme to work a little faster. If that happened in real life, your body could break down Lunesta a bit quicker, which in theory might make it less effective, so you could have a harder time falling or staying asleep.

Here's the reassuring part: this is rated a minor interaction, and the evidence comes only from test-tube and animal studies, not from people. The effect seen was far weaker than known enzyme boosters. I wouldn't lose sleep over it, but since cascara isn't meant for long-term use anyway, check with your pharmacist or doctor before pairing the two.

Cascara sagrada is a stimulant laxative made from the aged bark of a Pacific Northwest tree, used mainly for short-term relief of constipation. Because it can cause cramping, dehydration, and electrolyte problems—and was removed from over-the-counter laxative products in the U.S. due to safety concerns—it should only be used briefly, if at all, and with guidance from a pharmacist or doctor.

What Is Eszopiclone Used For?

Brand name: Lunesta

Eszopiclone tablets — sold under the brand name Lunesta and as generics — are indicated for the treatment of insomnia. In clinical studies, eszopiclone helped people fall asleep more quickly (reduced sleep latency) and stay asleep through the night (improved sleep maintenance).

Clinical trials supporting its effectiveness lasted up to 6 months in adults and up to 2 weeks in elderly patients. Lunesta and eszopiclone generics are approved for this same indication across the available tablet formulations.
