Leniolisib falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Devil's Claw interacts with.

Theoretically, devil's claw might increase levels of drugs metabolized by CYP3A4.
In vitro research shows that devil's claw might inhibit CYP3A4, although this has not been reported in humans.

Here's the short version: your body clears leniolisib (Joenja) using a liver enzyme called CYP3A4. Devil's claw may slow that same enzyme down. If it does, leniolisib could clear out more slowly and build up a bit higher than expected, which in theory might raise the chance of side effects. Leniolisib is an active drug, not a prodrug, so a blocker like this would tend to increase its effect rather than weaken it.

I want to be honest about how thin the evidence is, though. This comes only from lab testing, not from people, so we don't actually know if it matters in real life. Since leniolisib treats a serious immune condition, don't change anything on your own. Just run devil's claw past your pharmacist or doctor first.

What Is Leniolisib Used For?

Brand name: Joenja

Joenja (leniolisib) is approved to treat activated phosphoinositide 3-kinase delta syndrome, commonly called APDS — a rare genetic immune disorder in which certain immune cells (B and T cells) don't work properly. It is approved for adults and pediatric patients who are at least 12 years old.

APDS is caused by gene variants that make a specific enzyme (PI3K-delta) overactive, leading to immune system dysfunction. Joenja is the brand name for leniolisib in tablet form, taken by mouth.
