Devil's Claw and Lofepramine

Here is the interaction we found between these two, with the evidence behind it.

Moderate

CYTOCHROME P450 2C19 (CYP2C19) SUBSTRATES

Why this applies to Lofepramine

Lofepramine falls under CYTOCHROME P450 2C19 (CYP2C19) SUBSTRATES — the group Devil's Claw interacts with.

Theoretically, devil's claw might increase levels of drugs metabolized by CYP2C19.
In vitro research shows that devil's claw might inhibit CYP2C19, although this has not been reported in humans.
Severity Moderate Likelihood Possible Evidence D

Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.

Dr. Brian Staiger, PharmD, BCPS
Dr. Brian Staiger explains PharmD, Clinical Pharmacist

Here's the basic idea with Lofepramine, an older tricyclic antidepressant. Your liver uses an enzyme called CYP2C19 to help break this medicine down. Lab tests suggest devil's claw might slow that enzyme a bit, which could let lofepramine build up slightly in your system. If that happened, you might notice more of the usual tricyclic side effects, like a dry mouth, drowsiness, constipation, blurry vision, or feeling lightheaded when you stand up.

That said, this is theoretical. It comes from test-tube research only and has not been shown to happen in real people. So I wouldn't panic, but I'd give your pharmacist or doctor a heads-up before adding a concentrated devil's claw supplement so they can keep an eye on how you feel.
