Theoretically, indole-3-carbinol might increase the metabolism of CYP1A2 substrates and lower serum concentrations.

Here's the basic idea: your liver uses an enzyme called CYP1A2 to help break down estradiol. Indole-3-carbinol (the compound from broccoli and cabbage) may speed that enzyme up. If it does, your body could clear estradiol a little faster, which might lower how much is working in your system. In practice that could mean your menopause symptoms (vaginal dryness, itching, burning, or hot flashes) aren't controlled as well as you'd expect.

I want to be honest about the strength of this, though. This concern comes from lab and animal studies only, not from people, so it's theoretical and may not amount to much in real life. Food amounts of broccoli are very unlikely to matter; concentrated supplement doses are the bigger question. Before adding an I3C supplement, just check with your pharmacist or doctor first.

Here's the gist of it: Indole-3-carbinol (I3C) may push back against the effects of your estradiol. I3C nudges the liver to break down estrogen toward different forms, and lab work suggests it has an anti-estrogen action overall. So in theory, layering an I3C supplement on top of your estradiol could blunt how well the estradiol works.

With vaginal estradiol especially, that could mean your dryness, itching, burning, or painful intercourse don't ease up as much as you'd expect. I want to be honest, though: this concern comes mostly from lab and animal studies, so it's not firmly proven in people. Everyday broccoli and cabbage are very unlikely to matter; concentrated I3C capsules are the real question. Before adding it, please check with your pharmacist or doctor first.
