Melatonin + Ropinirole
Ropinirole brand names: Requip, Adartrel.
Melatonin has a moderate interaction with Ropinirole. Theoretically, melatonin might increase levels of drugs metabolized by CYP1A2.

Here is the interaction we found between these two, with the evidence behind it.
CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES
Why this applies to Ropinirole
Ropinirole falls under CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES — the group Melatonin interacts with.
Melatonin is metabolized in the liver primarily by the CYP2C19 and CYP1A2 enzymes. Theoretically, combined administration of melatonin with drugs metabolized by the CYP1A2 enzyme might reduce the metabolism of these drugs, resulting in increased serum levels. Conversely, some drugs metabolized by CYP1A2 may inhibit the metabolism of melatonin, resulting in increased serum levels of melatonin. Until more is known, use melatonin cautiously in patients taking drugs metabolized by these enzymes.
Interaction has not been documented in well-controlled studies, however, the interaction has been demonstrated in some small human studies or in controlled animal studies in conjunction with multiple case reports..
Here's the gist for your situation: your body clears ropinirole using a liver enzyme called CYP1A2, and melatonin may gently slow that same enzyme down. In theory, that could let a little more ropinirole build up than usual. Ropinirole is the active drug (not a prodrug), so higher levels generally mean stronger effects and a greater chance of side effects like daytime drowsiness, dizziness, lightheadedness when standing up, or nausea.
I want to keep this in perspective though. This concern is theoretical, based on how the two are processed rather than real-world reports, and many people use them together without trouble. Since both can also make you sleepy, just be aware of that overlap. Before starting melatonin, give your pharmacist or doctor a quick heads-up so they can keep an eye on how you feel.

Clinically significant. Avoid the combination, or use only under close professional supervision.
Usually worth avoiding or managing — may need monitoring, a dose change, or timing apart.
Generally not serious. Be aware of it and mention it to your pharmacist.

By severity: 1 major · 67 moderate · 33 minor.
From our pharmacist’s overview on the Ropinirole page
“Activated charcoal deserves the closest look on this page. It is the only one of the 101 entries rated major against ropinirole (Requip, Adartrel), because charcoal can bind the drug in the gut and shrink the amount that actually reaches you, and for someone relying on ropinirole for Parkinson's disease or restless legs, that can mean…”

About Melatonin
What is Melatonin?
- Trouble falling asleep (insomnia)
- Jet lag
- Delayed sleep phase syndrome
- Non-24-hour sleep-wake disorder
- Shift-work sleep problems
- Sleep problems in some children with conditions like autism or ADHD
Melatonin is a hormone your body makes naturally to help control your sleep-wake cycle, and the supplement form is widely used to help with sleep timing problems and jet lag. The evidence is strongest for certain circadian (body-clock) disorders and is more mixed for everyday insomnia. It is generally well tolerated for short-term use, but talk with your pharmacist or doctor before starting, especially for children or during pregnancy.
About Ropinirole
What Is Ropinirole Used For?
Brand names: Requip, Adartrel
Ropinirole tablets are used to treat two conditions: Parkinson's disease and moderate-to-severe primary Restless Legs Syndrome (RLS) — the uncomfortable urge to move the legs, especially at night or at rest.
Ropinirole extended-release tablets are indicated specifically for Parkinson's disease only. If you take the extended-release form, it is not approved for RLS — that use applies to the regular (immediate-release) tablets.
