Here's the gist for your NyQuil and adrafinil question. Once your liver turns adrafinil into modafinil, that modafinil may slightly speed up a liver enzyme called CYP1A2. In theory, that could nudge the breakdown of certain drugs a little faster. Of the ingredients in NyQuil, acetaminophen is the one with any real CYP1A2 link, so it could be cleared somewhat quicker.

Honestly, this is a theoretical concern based on lab data, not real-world reports, and the effect is described as weak. I wouldn't expect you to notice much. The bigger caution is that mixing a stimulant like modafinil with the sedating part of NyQuil (doxylamine) sends mixed signals to your body. Adrafinil also stresses the liver, and so can acetaminophen, so please run this combo by your pharmacist first.

Here's the gist with NyQuil and adrafinil. Once your liver turns adrafinil into modafinil, that modafinil can mildly speed up a liver enzyme called CYP3A4. A couple of NyQuil's ingredients, like dextromethorphan (the cough suppressant), lean partly on that same enzyme to be broken down. So in theory, clearing them a bit faster could make the cough and cold relief feel slightly weaker than usual.

Honestly, this is a theoretical concern based on how the drugs work, not something we see causing real problems in practice, and the enzyme effect is weak. The bigger reason to be cautious is liver strain, since both adrafinil and acetaminophen are processed by the liver. Please run this combo by your pharmacist or doctor first.

Here's the piece that matters with NyQuil: it contains pseudoephedrine, a decongestant that's also a mild stimulant, and it can speed up your heart and raise your blood pressure a bit. Adrafinil turns into modafinil in your body, which has its own wakefulness-promoting, stimulant-like effects. Stack the two and those effects can add up.

In real terms, you might notice a racing or pounding heartbeat, jitteriness, trouble sleeping, headache, or higher blood pressure. This is a theoretical concern based on how both work, not something proven in studies, so don't panic if you've combined them. Still, since adrafinil is unregulated and hard on the liver, it's worth checking with your pharmacist or doctor before pairing them, especially if you have any heart or blood pressure issues.
