Bazedoxifene Acetate, Conjugated Estrogens falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Adrafinil interacts with.

Adrafinil is a precursor to the stimulant prescription drug modafinil. Modafinil has been shown to weakly induce CYP3A4 activity.

Here's the situation with Duavee (conjugated estrogens plus bazedoxifene). Both the estrogen and bazedoxifene parts are processed by a liver enzyme called CYP3A4. When adrafinil gets converted into modafinil in your body, modafinil can mildly speed up that same enzyme. In theory, that means your body could clear Duavee a little faster than usual.

What might that look like? Possibly weaker results from Duavee, such as your hot flashes creeping back, or less protection for your bones over time. I want to be honest though: this is a theoretical concern based on how these drugs work, not something proven in real-world studies, and modafinil is only a weak booster of this enzyme. Still, since adrafinil is unregulated and hard on the liver, please run it by your doctor or pharmacist before combining them.

What Is Estrogen and Bazedoxifene Used For?

  • Estrogen and bazedoxifene tablets are used to treat hot flashes (sudden feelings of warmth, especially in the face, neck, and chest) in women who are experiencing menopause (stage of life when menstrual periods become less frequent and stop and women may experience other symptoms and body changes). Estrogen and bazedoxifene tablets are also used to prevent osteoporosis (condition in which the bones become thin and weak and break easily) in women who have undergone menopause. Estrogen is in a class of medications called hormones and bazedoxifene is in a class of medications called estrogen agonist–antagonists. Estrogen works by replacing estrogen that is normally produced by the body. Bazedoxifene is used to block the action of estrogen on the lining of the uterus, decreasing the risk of overgrowth that may lead to cancer.
