Here's the thing about Auvelity (the bupropion plus dextromethorphan combo used for depression): the bupropion piece is there partly to block an enzyme so the dextromethorphan sticks around longer in your system. Adrafinil turns into modafinil in your liver, and modafinil can mildly speed up a different enzyme called CYP3A4. In theory, that faster clearance could nudge the levels of this medication down a bit, possibly making it work less well for your mood.

I'll be honest with you, this is a theoretical concern based on how these drugs behave in the lab, not something we've seen cause real problems in people. The effect on CYP3A4 is described as weak. Still, before pairing an unregulated supplement like adrafinil with Auvelity, please check with your pharmacist or prescriber.

Auvelity combines bupropion and dextromethorphan. Here, the bupropion piece is the part that matters. Your liver uses an enzyme called CYP2B6 to break bupropion down. Adrafinil turns into modafinil in your body, and in lab studies modafinil seems to mildly speed up that enzyme. If that happened in real life, your body might clear the bupropion a little faster, which could slightly weaken the antidepressant effect over time.

I want to be honest: this is theoretical, based only on test-tube data, so the real-world impact is probably small. Still, since Auvelity is a depression treatment, it is worth mentioning to your pharmacist or doctor before mixing it with adrafinil, especially since adrafinil itself is unregulated and hard on the liver.
