Theoretically, adrafinil might reduce the levels and clinical effects of CYP1A2 substrates.
Adrafinil is a precursor to the stimulant prescription drug modafinil. Modafinil has been shown to weakly induce CYP1A2 activity in vitro.

Here's the gist for Zofran (ondansetron): your body partly clears it using a liver enzyme called CYP1A2. Adrafinil, after your liver turns it into modafinil, may gently speed up that enzyme. In theory, that could break down ondansetron a little faster and lower how much is in your blood, which might make it slightly less effective at keeping nausea and vomiting away.

I want to be straight with you, though: this is a theoretical concern based on how the drugs behave in lab studies, not real patient reports, and the effect on the enzyme looks weak. It's worth mentioning to your pharmacist or doctor, especially if your nausea isn't well controlled, but it's not a reason to panic.

About Ondansetron

What Is Ondansetron Used For?

Brand name: Zofran

Ondansetron is used to prevent nausea and vomiting in several situations. Oral forms (tablets, orally disintegrating tablets, and oral solution) are approved to prevent nausea and vomiting from highly or moderately emetogenic (nausea-causing) cancer chemotherapy, from radiation therapy to the abdomen or whole body, and from surgery.

Ondansetron injection (given intravenously or intramuscularly in a clinical setting) is approved to prevent nausea and vomiting from emetogenic cancer chemotherapy — including high-dose cisplatin — in adults and children as young as 6 months old, as well as to prevent postoperative nausea and vomiting.
