Here's the gist of it. Once you take adrafinil, your liver converts it into modafinil, and modafinil is cleared from your body largely by an enzyme called CYP3A4. Ritonavir (Norvir) is a strong blocker of that same enzyme. So in theory, ritonavir could slow the breakdown of modafinil and let it build up, which may push the wakefulness effects too far. That could mean trouble sleeping, jitteriness, a faster or pounding heartbeat, anxiety, or headaches.

I want to be honest that this is a theoretical concern based on how the drugs work, not from solid studies. Still, since adrafinil is unregulated and can stress the liver, I'd run it by your doctor or pharmacist before combining the two, rather than guessing.

Here's the idea with adrafinil and ritonavir (Norvir). Once your liver turns adrafinil into modafinil, that modafinil can gently speed up an enzyme called CYP3A4. Ritonavir is partly handled by that same enzyme, so in theory your body could clear it a little faster and leave somewhat lower levels in your blood.

Why that matters: ritonavir is often used to keep HIV under control or to boost other HIV medicines. Lower levels could mean less of that effect over time. I want to be honest though, this is a theoretical concern based on how the drugs work, not strong human studies, and modafinil's effect on this enzyme is weak. Since HIV treatment is so important, please check with your doctor or pharmacist before combining them.

What Is Ritonavir Used For?

Brand name: Norvir

Ritonavir (brand name Norvir) is used together with other HIV medicines to treat HIV-1 infection in adults and children older than one month of age. It is always taken as part of a combination regimen — never on its own.

Norvir oral powder is specifically indicated for pediatric patients with HIV-1 infection. Norvir tablets and oral solution are indicated for both adults and pediatric patients with HIV-1 infection. Ritonavir tablets (generic) are also indicated for the treatment of HIV-1 infection in combination with other antiretroviral agents.
