Ashwagandha + Amitriptyline, Chlordiazepoxide
Amitriptyline, Chlordiazepoxide brand name: Limbitrol DS.
Amitriptyline, Chlordiazepoxide falls into 5 drug categories that Ashwagandha can interact with; the most serious is rated moderate. Theoretically, taking ashwagandha might increase the sedative effects of CNS depressants.

These two interact in 5 distinct ways. Each one is explained separately below.
Why Are There 5 Interactions?
Ashwagandha interacts with whole categories of medication. Amitriptyline, Chlordiazepoxide falls into 5 of those categories, so it can interact with Ashwagandha in 5 different ways. We’ve listed each one below — your overall risk is driven by the most serious of them (Moderate).
CNS DEPRESSANTS
1 of 5Why this applies to Amitriptyline, Chlordiazepoxide
Amitriptyline, Chlordiazepoxide falls under CNS DEPRESSANTS — the group Ashwagandha interacts with.
Ashwagandha seems to have sedative effects. Theoretically, this may potentiate the effects of barbiturates, other sedatives, and anxiolytics.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Limbitrol combines two medicines that already calm the brain: amitriptyline (a sedating antidepressant) and chlordiazepoxide (a benzodiazepine in the same family as Valium). Both can leave you feeling drowsy, foggy, or unsteady on their own. Ashwagandha is thought to have mild calming, sleep-promoting effects of its own, so the worry here is simply that stacking calm on top of calm could deepen those effects.
In plain terms, you might feel more sleepy, slowed-down, or dizzy than usual, which raises the risk of falls or trouble concentrating. To be honest, this caution is theoretical and based on how the herb is believed to act, not on strong human studies. Still, since this combo is already heavily sedating, it is worth a quick chat with your pharmacist or doctor before adding ashwagandha.

BENZODIAZEPINES
2 of 5Amitriptyline, Chlordiazepoxide also falls under BENZODIAZEPINES.
There is preliminary evidence that ashwagandha might have an additive effect with diazepam (Valium) and clonazepam (Klonopin). This may also occur with other benzodiazepines.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Limbitrol DS combines two calming medicines: amitriptyline (a sedating antidepressant) and chlordiazepoxide (a benzodiazepine). Both already tend to make people feel drowsy and relaxed. Ashwagandha is often taken to ease stress and help with sleep, so the concern here is simply that the calming effects could pile up. In theory, the combination might leave you feeling more sleepy, foggy, dizzy, or slowed down than usual.
I want to be honest that this is a theoretical caution based on how these things work, not strong human studies, so don't panic if you've taken them together. Still, since both already cause drowsiness, it's worth a quick chat with your pharmacist or doctor before adding ashwagandha, especially before driving or at bedtime.

CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES
3 of 5Amitriptyline, Chlordiazepoxide also falls under CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES.
In vitro research shows that ashwagandha extract induces CYP1A2 enzymes.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Limbitrol DS combines amitriptyline (an antidepressant) with chlordiazepoxide (a calming benzodiazepine). Neither one is a prodrug, so they work as-is once you take them. There's a lab-based idea that ashwagandha could nudge up an enzyme called CYP1A2, which helps clear amitriptyline. If that happened in real life, it could lower amitriptyline levels a bit and make it work a little less, meaning your mood or sleep benefit might slip.
Honestly, this is theoretical and only seen in test-tube studies, not in people, so I wouldn't lose sleep over it. The bigger real-world concern is that ashwagandha can add to drowsiness from this combo. If you're considering it, just run it by your pharmacist or doctor first.

CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES
4 of 5Amitriptyline, Chlordiazepoxide also falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES.
In vitro research shows that ashwagandha extract induces CYP3A4 enzymes.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Limbitrol combines amitriptyline (an antidepressant) with chlordiazepoxide (a sedative from the same family as Valium). Both are processed in part by a liver enzyme called CYP3A4. Some early lab research hints that ashwagandha may speed that enzyme up, which could, in theory, clear these medicines a little faster and make them work less well. For amitriptyline that might mean less help with mood or nerve pain, and for chlordiazepoxide possibly less calming effect.
Honestly, this is a minor concern based only on test-tube data, so I wouldn't lose sleep over it. Still, since both drugs already cause drowsiness, it's worth a quick chat with your pharmacist or doctor before adding ashwagandha so they can keep an eye on how you respond.

Serotonergic Drugs
5 of 5Amitriptyline, Chlordiazepoxide also falls under Serotonergic Drugs.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Limbitrol DS pairs amitriptyline, an older antidepressant that raises serotonin, with chlordiazepoxide, a calming benzodiazepine. The thinking here is that ashwagandha may gently nudge serotonin activity too, so combining the two could in theory add up. In real life this concern is small. It comes mostly from animal research, and there are no published reports of serotonin syndrome from this combo.
What I'd watch for instead is the calming side adding up. Ashwagandha is often taken for sleep and stress, and both parts of Limbitrol can already make you drowsy. So you might feel extra sleepy, foggy, or dizzy. Don't start or stop anything on your own. Just check with your pharmacist or doctor first, especially if you also have thyroid or liver issues.

From our pharmacist’s overview on the Amitriptyline, Chlordiazepoxide page
“Alcohol is the first thing to know about here. Beer and wine both carry major, well-documented interactions with Amitriptyline and Chlordiazepoxide (Limbitrol DS), because both drugs already slow the central nervous system, and anything sedating layered on top can deepen ordinary drowsiness into something dangerous.”
