Agomelatine falls under CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES — the group Bacopa interacts with.
Research on the effects of bacopa extracts on CYP1A2 enzymes is conflicting. Some in vitro evidence shows that bacopa extract can moderately and non-competitively inhibit CYP1A2, while other in vitro evidence suggests that any effect is unlikely to be clinically significant.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the situation with bacopa and Valdoxan (agomelatine). Your body relies heavily on a liver enzyme called CYP1A2 to clear agomelatine. In lab dishes, bacopa might slow that enzyme down a little. If it did, agomelatine could build up more than usual, which in theory could mean more of its typical effects like daytime sleepiness, dizziness, headache, or nausea.
I want to be honest though: this is only a theoretical concern based on test-tube work, and the studies even disagree with each other. We don't have real human evidence that it causes problems. Still, since agomelatine already needs liver monitoring, it's worth a quick mention to your doctor or pharmacist before adding bacopa.

In vitro evidence suggests that bacopa extract can moderately and non-competitively inhibit CYP2C19 enzymes. It is not known whether this is clinically significant.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the simple version. Your body relies on liver enzymes to break down agomelatine (Valdoxan), and one of those enzymes is called CYP2C19. In test-tube studies, bacopa appears to mildly slow that enzyme down. If that happened in a real person, agomelatine could clear a bit more slowly and build up, which might mean stronger drowsiness, dizziness, headache, or nausea.
The honest part: this is only laboratory evidence right now, so we truly don't know if it matters in everyday life. I wouldn't lose sleep over it, but since agomelatine already needs liver monitoring, it's worth a quick chat with your pharmacist or doctor before adding a daily bacopa supplement so they can keep an eye on how you feel.

Research on the effect of bacopa extracts on CYP2C9 enzymes is conflicting. Some in vitro evidence suggests that bacopa extract can moderately and non-competitively inhibit CYP2C9, while other in vitro evidence suggests that any effect is unlikely to be clinically significant.

Here's the gist: in some lab studies, bacopa may slow down a liver enzyme called CYP2C9. Agomelatine is partly broken down by that pathway, so in theory bacopa could let a little more of it linger in your body. Agomelatine is not a prodrug, so more drug hanging around generally means a stronger effect and a bit more chance of side effects like daytime sleepiness, headache, dizziness, or nausea.
That said, I want to keep this in perspective. This concern comes only from test-tube research, and even those studies disagree on whether it matters in real life. We mostly watch the liver with agomelatine anyway, so it's worth a quick chat with your pharmacist or doctor before combining them, but there's no need to panic.

Bacopa is an Ayurvedic herb most often used for memory and thinking. Some small studies suggest it may modestly help memory when taken regularly for several weeks, but the evidence is limited and quality varies.
