Amitriptyline falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Breadfruit interacts with.
Some drugs metabolized by CYP3A4 include amitriptyline (Elavil), amiodarone (Cordarone), citalopram (Celexa), felodipine (Plendil), lansoprazole (Prevacid), ondansetron (Zofran), prednisone (Deltasone), sertraline (Zoloft), and many others.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the gist: your body uses a liver enzyme called CYP3A4 to help break down amitriptyline. In lab and animal studies, certain compounds in breadfruit leaves seem to slow that enzyme down a bit. If that happened in people, amitriptyline could clear a little more slowly and build up, which might mean more of its usual side effects like drowsiness, dry mouth, constipation, blurry vision, or feeling lightheaded when you stand up.
The honest part: this is only a theoretical concern based on test-tube and animal data, and eating breadfruit as a food is very unlikely to cause a problem. Concentrated leaf teas or extracts are the bigger question mark. If you're using a breadfruit product for a health reason, just run it by your pharmacist or doctor first.
