Amitriptyline falls under TRICYCLIC ANTIDEPRESSANTS (TCAs) — the group Cowhage interacts with.

Theoretically, use of TCAs might reduce the levels and clinical effects of cowhage.
Cowhage contains levodopa. Use of TCAs might reduce the absorption of levodopa. Some case reports describe patients that developed hypertension and dyskinesia when taking both levodopa and TCAs.

Here's the gist: cowhage works because it's packed with L-dopa, the natural form of the Parkinson's drug levodopa. When you pair it with amitriptyline (Elavil), the tricyclic can slow how your gut absorbs that L-dopa, so the cowhage may simply work less well than expected. On top of that, a few older case reports noted people getting spikes in blood pressure and unwanted muscle movements (dyskinesia) when combining levodopa with a tricyclic.

I want to be honest that this is theoretical, based mostly on how these compounds behave rather than strong studies. Still, both have real effects on dopamine and blood pressure, so it's worth a quick chat with your doctor or pharmacist before mixing them, especially since cowhage dosing is unpredictable.
