Bosentan falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Dhea interacts with.

Theoretically, DHEA might increase the levels of drugs metabolized by CYP3A4.
Some preliminary evidence shows that DHEA may inhibit CYP3A4; however, the clinical significance of this potential interaction is not known.

Here's the gist with DHEA and Tracleer (bosentan). Your liver uses an enzyme called CYP3A4 to help clear bosentan from your body. There's some early, fairly thin evidence that DHEA might slow that enzyme down a little. If it does, bosentan could build up more than usual, which in theory might mean more of its typical side effects, things like headache, flushing, swelling in the legs or ankles, low blood pressure, or feeling unusually tired.

I want to be honest though: this is mostly theoretical and based on weak, anecdotal reports, so we don't really know if it matters in real life. Bosentan is a medicine that already needs close monitoring, so before adding DHEA, please run it by your pharmacist or doctor first.

What Is Bosentan Used For?

Brand name: Tracleer

Bosentan is used to treat pulmonary arterial hypertension (PAH) — high blood pressure in the arteries of the lungs. In adults, it helps improve the ability to exercise and slows how quickly the disease gets worse.

In children aged 3 years and older with idiopathic (no known cause) or congenital (present from birth) PAH, bosentan is used to lower resistance in the lung blood vessels, which is expected to improve exercise ability. Tracleer is one brand approved for both adults and pediatric patients with PAH.
