Here is the interaction we found between these two, with the evidence behind it.
CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES
Why this applies to Darunavir, Ritonavir
Darunavir, Ritonavir falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Jequirity interacts with.
Interaction has been demonstrated in animal or in lab research but has been shown not to occur in humans..
First, the most important point about jequirity has nothing to do with metabolism: the seeds contain abrin, one of the deadliest plant toxins we know of, similar to ricin. This is not something anyone should be taking as a supplement, full stop.
On the interaction itself, the concern is theoretical and comes only from lab (test tube) work, where jequirity extract slowed the CYP3A4 enzyme. Your darunavir/ritonavir combo already leans on this same pathway (ritonavir intentionally blocks it to boost darunavir). In theory, an added blocker could nudge drug levels higher, meaning more nausea, diarrhea, or headache. In real life, this is unlikely and unproven. Please don't use jequirity, and check with your pharmacist or HIV provider before adding anything new.
