Here's the simple version: stimulants like amphetamine sulfate (Evekeo, Benzedrine) can slightly lower the seizure threshold, meaning they make seizures a little easier to trigger in people who are already prone to them. There's some thought that melatonin, in certain people, might do the same thing. Put the two together and the concern is a small, mostly theoretical bump in seizure risk.

I want to be honest about how soft this evidence is. It's rated theoretical, based mostly on how these things work, with a little real-world signal in children who have neurological conditions. For most healthy adults using melatonin short-term for sleep, this is unlikely to be a real problem. If you or your child has a seizure history, though, please run it by your pharmacist or doctor first.

CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES

Amphetamine Sulfate also falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES.

Theoretically, melatonin might increase levels of drugs metabolized by CYP2D6.
Laboratory research suggests that certain lots of melatonin inhibit CYP2D6. Theoretically, combined administration of melatonin with certain drugs metabolized by the CYP2D6 enzyme may reduce the metabolism of these drugs, resulting in increased serum levels. Until more is known, use melatonin cautiously in patients taking drugs metabolized by these enzymes.

Here's the gist if you take melatonin to help you sleep while you're on amphetamine sulfate (Evekeo, Benzedrine). Your liver uses an enzyme called CYP2D6 to help clear amphetamine. In lab studies, some batches of melatonin seemed to slow that enzyme down a bit. In theory, that could let a little more amphetamine linger in your system, which might mean stronger stimulant effects like a faster heartbeat, jitteriness, trouble winding down, or higher blood pressure.

The reassuring part: this is rated minor and comes only from test-tube research, not real patients. For most people it's unlikely to cause a noticeable problem. Still, since melatonin is for sleep and amphetamine is a stimulant, run the combo by your pharmacist or doctor first.
